Continuing problems...

 

Yesterday I put 2 of the 4 plates into the 37 degree incubator to see if it was a growth time issue. Both plates had growth, but not necessarily E. coli. The one plate was a mold or fungus of some sort, it looked like. The other had very large colonies that could be E. coli, but not definatively. They could have been something else as well.

 

The plate that Crystal streaked showed no visible growth.

 

We decided to leave it for another day and see if anything grows up, it might just be that overnight is not nearly enough at room temperature.

 

As a note, neither Allison nor Crystal have ever used a competence procedure quite like this. Looking through some of the references, I'm not sure why it's the recommended protocol. We may need to investigate one of the other protocols if this doesn't turn out. I photocopied and checked out some stuff from the library that we should look at.
