Lab Sept 08 2009

So I was thinking that tomorrow (Wednesday the 9th) might be a good time for all of us who're available to come up to the new lab and check things out, maybe mix up some medium and/or subculture some wee beasties. I'm available from about 1:00pm to 5:15pm (need some lunch after my classes). How about everyone else?

 

Natasha, 11.15am:

Is anyone else coming in today?  Also, did we ever talk to anyone about freezer boxes, or should I do that today?  I can be in tomorrow after 12.30.

 

12.20pm

Alright, our -80C stocks are in their new freezer; we still need boxes though.  I've talked with Melissa and Barb and they're happy with state of the old lab.  We still need to figure out what we're doing for glassware etc.

 

2.37pm

We have started some new assemblies today:

K235009, 13, 14, 16 and 17.  (We would have done 15, but R0051 is still inconsistent.) 

So new U I0500, D J23032, and DP P1010 Kan.  We also did a new DP digestion of P1010 Cm because we were running low.

 

 To save on competent cells we will do all of the transformations tomorrow as the Kan ones can't be done till we have new plates.

 

Tomorrow (Wed):

- New plates

- Transform parts

 

Thurs:

- Broth culture parts

 

Fri:

- Miniprep parts

- Gel

 

4:15pm

 

Before I (Kyliah) left, I subcultured the R0051 in about 75mL LB and left it in the shaker thing in the corner. Be careful, it starts beeping up a storm if the temperature drops due to lid open.
